John Shaft, un policier new-yorkais, arrête Walter Wade Jr, un étudiant en cheville avec un trafiquant de drogue. Il l'accuse d'avoir assassiné un camarade. Libéré sous caution, le jeune homme s'enfuit en Suisse. Deux ans plus tard, alors qu'il est rentré aux Etats-Unis clandestinement, il est à nouveau coincé par Shaft. Cette fois-ci, pas question pour lui de disparaître. L'étudiant, confiant en l'influence et en la fortune de son père, décide d'affronter la justice. Mais il lui faut d'abord faire taire à jamais un embarrassant témoin oculaire. Shaft doit absolument retrouver ce dernier et le protéger des hommes de main du jeune assassin...
